Our objectives were to determine the safety and hemodynamic response of Adenosine Triphosphate-Magnesium Chloride (ATP-MGCL2) infusion in normal awake human volunteers. Five healthy adult male volunteers received an intravenous infusion of ATP-MGCL2 (0.1-0.4mg/kg/min) on four separate occasions. The totals dose infusion was 3,6,10 and 30mg/kg. Hemodynamic measurements were made at end exhalation in the supine position and included heart rate and systolic, diastolic and mean blood pressure. Continuous electrocardiographic monitoring ...

The purpose of our study was to determine how ATP-Magnesium Chloride might be used, what the potential problems with ATP-MGCL2 might be and to develop all the necessary background information in order to initiate clinical trials. In normal as well as hypovelemic awake dogs, cardiac output can be increased significantly by infusion of ATP-MGL2 intravenously at rates of 0.5-2. 5 mg/Kg/min. Although higher doses of S4ATP-MgL2 may have detrimental hemodynamic ...

The purpose of our studies was to determine the safety and hemodynamic response of adenosine triphosphate/magnesium chloride infusion in normal awake human volunteers. In accordance with a protocol approved by the Human Investigation Committee, five healthy adult male volunteers received an intravenous infusion of adenosine triphosphate/magnesium chloride (o.1-0.4mg/kg/ min) on four separate occasions. The total dose infused was 3, 6, 10 and 30mg/kg (n=20 studies). Hemodynamic measurements were made at ...

In conscious rate and primates, bolus infusion of the entire efficacious dose of ATP-MgCl2 transiently decrease mean blood pressure but there were no long term side effects and there were no mortalities. Moreover, daily administration of the entire efficacious dose of ATP-MgCl2 for a period of three months in rats did not produce any mortality or adverse side effects. ATP-MgCl2 was approved for clinical studies by the Food and Drug ...

A scheme of the progressive changes that occur with shock and ischemia which we have previously discussed in detail is presented in Figure 1. The major changes during such conditions are in microcirculation, cell membrane transport and function, energy metabolism and mitochondrial function. These progressive and interrelated events eventually lead to cell swelling. Studies showed that infusion of ATP-MgCl2 (along with volume restitution) following hemorrhagic shock and sepsis-peritonitis in animals ...
