The invasion plasmid antigens (Ipa) of Shigella spp, are effector molecules necessary for the invasion of colonic epithelial cells. Recently, we have isolated an invasin protein-LPS complex from intact, virulent Shigella (Invaplex). In vitro observations suggest that Invaplex interacts with host-cell membranes, is internalized, and is released into the cytoplasm via a process similar to the activity expressed by virulent shigellae. It was therefore hypothesized that if heterologous molecules (DNA ...

Tn5-tagged invasion plasmid DNA (pWR110) from Shigella flexneri serotype 5 (strain M90T) was cloned into the expression vector Lambda gt11. Recombinant phage (Lambda gt11Sfl) expressing pWR110-encoded polypeptide antigens were identified by using rabbit antisera directed against S.flexneri M90T invasion plasmid antigens. Antigens encoded by Lambda gt11Sfl recombinant phage were characterized by reacting affinity-purified antibodies, eluted from nitrocellulose-bound plaques of Lambda gt11Sfl recombinants, with virulent, wild-type S. flexneri M90T polypeptides in ...

Several polypeptide antigens of Rickettsia tsutsugamushi are recognized by human or primate convalescent sera and may be important protective immunogens. Molecular cloning and expression of the genes encoding the 110K (110 kilodalton) and 56K polypeptide antigens of R. tsutsugamushi Karp were accomplished in the lambda gt11 expression vector system. Southern blot analysis with the cloned fragments for the 56K polypeptide antigen (0.7 kilobases) and the 110K polypeptide antigen (5.4 kilobases) ...

This invention relates to nucleic acid probes and a method for the rapid detection of Shigella and enteroinvasive Escherichia coli, the causative agents of bacterial dysentery, by use of a nucleic acid hybridization probe, equivalent to a plasmid DNA region encoding one of 4 specific invasion-associated, peptides of all strains of Shigella and enterinvasive E. coli, in a nucleic acid hybridization reaction with a clinical specimen containing dysentery bacteria.
